Breast cancer accounts for 27.7% of newly diagnosed cancer cases in women worldwide, making it one of the leading causes of mortality in this group. This highlights the urgent need for less invasive and more effective therapies. In this context, Lentinula edodes commercially called Shiitake, has demonstrated antitumor, immunoregulatory, and antioxidant properties due to its rich composition of polysaccharides, amino acids, vitamins, lentinan, ergothioneine and ergosterol. This study evaluated the anticancer effect of the steam-distilled extract of L. edodes in an in vitro model, using triple-negative breast cancer cells (MDA-MB-231) and normal pulmonary fibroblasts (MRC-5) as controls. The results showed a significant reduction in the viability of MDA-MB-231 cells, with an LD50 of 40 %. In cell migration assays, treatment with the extract reduced tumor cell migration by 62% compared to the control. Additionally, a three-dimensional (3D) model was implemented to simulate the tumor microenvironment, where treated cells showed decrease in spheroid diameter and number after 5 days of treatment. Although these findings are promising, further studies are needed to analyze specific fractions of the extracts, utilize more complex 3D models, and conduct in vivo research to validate their clinical application. This work provides preliminary evidence supporting the use of natural products in oncology, opening new possibilities for integrative and personalized therapies.spa
